The purification of biological therapeutics requires processes which usually employ two technologies used in combination: membrane separations and chromatography. Chromatography offers greater selectivity than membrane separations, and can purify much greater quantities of solution in one operation.

However, membrane separations technologies offer a number of other advantages including the fact that they are a fast and robust process step.


ÄKTAcrossflow covers three main filtration techniques:

  • Ultrafiltration of proteins
  • Diafiltration of proteins
  • Microfiltration of cell and lysate solutions


The system supports both hollow fibers and flat sheet cassettes with sizes suitable for flow rates up to 300 ml/min for flat sheet cassettes and up to 600 ml/ min for hollow fibers.

The Typhoon FLA 7000 is a fast and versatile laser scanner for biomolecular imaging applications including sensitive and quantitative measurements of radioisotopic labels by storage phosphor and single fluorescence, as well as digitization of colorimetric stains (e.g., Coomassie™ Blue and silver-stained gels). The Ettan IPGphor II from GE Healthcare is the ideal option for obtaining perfectly resolved first dimensions of 2D gels. The manufacturer claims that the system provides ‘reproducibility, reliability, speed, accuracy, high protein loading capacity, and high throughput’, and in my experience, the system really stands up to their claim. To begin with, the system is very compact and can be housed on a tabletop in any corner of the lab. It is perfectly silent and occupies little space. The system requires an ambient, dust-free operating environment; however, we keep the instrument in a well air-conditioned room. The system contains gold-plated copper electrodes that provide the necessary contact to the 1D strips for carrying out isoelectric focusing. A very high voltage is applied between the electrodes and, therefore, caution should be used while handling the instrument. As a safety feature, the electrodes are automatically switched off when anyone opens the lid of the instrument.
